Near complete reference genome assembly of Corallodiscus lanuginosus, a widely distributed medicinal resurrection plant

Corallodiscus is an oligotypic genus comprising five species, with C. lanuginosus being the most widespread distributed species and characterized by its extensive morphological variation, high genetic diversity, and ecological and medicinal significance as a resurrection plant. These features make it a valuable model for investigating taxonomy, genome evolution, and desiccation tolerance. However, genomic studies of this species have been limited by the absence of a high-quality reference genome. Here, we present a near-complete genome assembly of C. lanuginosus , representing the first genomic resource for the subtribe Corallodiscinae, generated using PacBio HiFi and Hi-C sequencing. The assembled genome spans 572.8 Mb, with a contig N50 of 41.0 Mb, and 555.9 Mb (97.1%) anchored onto ten pseudochromosomes. The assembly achieves high continuity and accurate, with a BUSCO completeness of 98.2%, a HiFi read mapping rate of 98.5%, an LAI of 24.7, and a QV of 53.8. A total of 20,600 protein-coding genes were predicted, of which 19,822 (96.2%) were functionally annotated. This high-quality genome provides a foundational resource for future studies on the evolutionary biology, adaptive mechanisms, and medicinally relevant traits of Corallodiscus .
